Is there a way to improvise a melody on the synth keyboard while listening to the drum loop on the tracker mini, as shown in this video, and then record that melody on the mini?

Sadly playing back something on the Tracker while sampling at the same time will not be possible. See: Allow Sampling During Playback of Audio

This was declined due to hardware limitations. I’d suggest to record this to a secondary device and then either import it or sample it to the tracker from there then.
